不同激素配比和AgNO3 对两种基因型大白菜子叶离体再生的影响

摘 要:

以 ‘08S555’ 和 ‘ 陕秋白 ’ 大白菜子叶为外植体, 研究了激素配比和 AgNO3 对不定芽再生的影响。结果表明:‘ 陕秋白 ’ 的再生率高于 ‘08S555’, 但出芽系数比 ‘08S555’ 低。在 6-BA 与 NAA 组合时,‘ 陕秋白 ’ 再生率最高, 为 68.33%, ‘08S555’ 为 28.33%; 再生系数分别为 1.09 和 2.81。在 TDZ 与 NAA 组合时, 二者不定芽的再生频率分别为 79.17% 和 45.83%; 再生 系数分别为 1.24 和 2.55。这说明与 6-BA 相比, TDZ 与 NAA 组合对两个大白菜子叶再生率的效果更好。AgNO3 与细胞分 裂素及生长素配合使用, 能提高 ‘ 陕秋白 ’ 子叶不定芽的再生频率。当 TDZ 浓度为 0.3 mg•L-1、NAA 为 0.5 mg•L-1 和 AgNO3为 6 mg•L-1 时, ‘ 陕秋白 ’ 再生率最高, 达到 87.50%。

Effects of Different Hormone Combinations and AgNO3 on in vitro Regeneration from Cotyledons of Two Genotypes of Heading Chinese Cabbage

Abstract:

Cotyledons of ‘08S555’ and ‘shanqiubai ’ were used as explants to study the effects on regeneration of adventitious buds so as to choose optimal hormone combination with AgNO3. The results showed that‘shanqiubai’ had higher regeneration and lower sprouting number of explants than ‘08S555’. In the 6-BA and NAA combinations, ‘shanqiubai’ got the highest regeneration rate of 68.33%, but ‘08S555’ was only 28.33%. Regeneration coefficients were 1.09 and 2.81. In TDZ and NAA combinations, 79.17% and 45.83% of adventitious bud in ‘shanqiubai’ and ‘08S555’ regenerated, with regeneration coefficients of 1.24 and 2.55, respectively. With 6-BA in comparision, TDZ was more effective than 6-BA. AgNO3 used in conjunction with the cytokinin and auxin can enhance cotyledon regeneration frequency of ‘shanqiubai’ adventitious buds. When 6-BA/TDZ combinated with NAA and AgNO3, the highest regeneration rate is 87.50% of ‘shanqiubai’, and the suitable concentration was TDZ (0.3 mg·L-1), NAA (0.5 mg·L-1) and AgNO3 (6 mg·L-1) separately.
